Abstract:
The CYP1A2 gene encodes the primary enzyme responsible for metabolizing caffeine,
and variation at the rs762551 SNP affects how quickly caffeine is processed. Fast
metabolizers (AA) break down caffeine rapidly, while slow metabolizers (AC or CC)
experience longer-lasting effects. The purpose of this project is to determine each
participant’s CYP1A2 rs762551 genotype using PCR-RFLP analysis and investigate how
genetic variation correlates with caffeine consumption habits and physiological responses.
Participants collected buccal cells to isolate genomic DNA, amplified the SNP region
via PCR, and performed RFLP analysis with Apal and Sacl restriction enzymes.
To connect genotype with caffeine behavior, participants completed a survey assessing
frequency and type of caffeine consumption, typical daily intake, perceived effects,
duration of effects, and impact on sleep. By comparing survey results with genotypes,
students can explore how genetics influences caffeine metabolism, tolerance, and sensitivity,
demonstrating the role of genetic variation in individual physiological responses.
